چکیده

Epidermal hyperplasia is a protective response to restore disrupted skin barrier. Innate lymphoid cells (ILCs) have recently emerged as crucial resident immune for tissue homeostasis; however, how ILCs regulate the barrier after mechanical damage under physiological conditions remains unclear. In this study, we found that by mild tape stripping (T/S) induced epidermal with increased expression of proteins in WT mice and Rag2−/− (no T B cells), but was significantly reduced Rag2−/−Il2rg−/− ILCs). single cell RNAseq EdU pulse experiments, keratinocyte proliferation T/S mice, suggesting ILC promoted restoration via epithelial proliferation. While IL-7-deficient ILC2/3) responded similarly IL-15 deficient ILC1) showed hyperplasia. Unexpectedly, ILC1 major source Amphiregulin (AREG) both murine human skin, deficiency AREG led significant reduction Interestingly, release ATP from explants, highly sensitive sensor visualized extracellular (eATP) primary cultured keratinocytes stimulation. Furthermore, eATP receptor inhibitors suppressed T/S. Finally, performed Raman microscope imaging investigate damage-induced changes oxidative stress-related Increased anti-oxidant Rag2−/−mice less prominent lacking ILC1. Indeed, DNA epidermis more accumulated Taken together, ILC1-derived plays role eATP-induced protection stress.

Type 3 innate lymphoid cells maintain intestinal epithelial stem cells after tissue damage

Disruption of the intestinal epithelial barrier allows bacterial translocation and predisposes to destructive inflammation. To ensure proper barrier composition, crypt-residing stem cells continuously proliferate and replenish all intestinal epithelial cells within days. As a consequence of this high mitotic activity, mucosal surfaces are frequently targeted by anticancer therapies, leading to ...

Innate lymphoid cells

Purpose of review: The purpose of this article is to review recent advances in our understanding of innate lymphoid cell function and to speculate on how these cells may become activated and influence the immune response to allogeneic tissues and cells following transplantation. Recent findings: Innate lymphoid cells encompass several novel cell types whose wide ranging roles in the immune syst...

Human innate lymphoid cells.

Innate lymphoid cells (ILCs) are lymphoid cells that do not express rearranged receptors and have important effector and regulatory functions in innate immunity and tissue remodeling. ILCs are categorized into 3 groups based on their distinct patterns of cytokine production and the requirement of particular transcription factors for their development and function. Group 1 ILCs (ILC1s) produce i...

HiJAKing Innate Lymphoid Cells?

The family of innate lymphoid cells (ILCs) consists of a heterogeneous group of cytokine-producing cells that have features in common with adaptive T helper (Th) cells. Cytokines acting through the Janus kinase (JAK)/signal transducer and activator of transcription (STAT) pathways are key players in both Th and ILC biology. Observations in animal models, supported by evidence from humans, have ...

Innate lymphoid cells promote anatomical containment of lymphoid-resident commensal bacteria.

The mammalian intestinal tract is colonized by trillions of beneficial commensal bacteria that are anatomically restricted to specific niches. However, the mechanisms that regulate anatomical containment remain unclear. Here, we show that interleukin-22 (IL-22)-producing innate lymphoid cells (ILCs) are present in intestinal tissues of healthy mammals. Depletion of ILCs resulted in peripheral d...
